Abstract Vitreoscilla hemoglobin (VHb) has been known to interact with various oxygenases and supply oxygen to oxygenase reactions. To investigate the possibility to generate engineered VHb’s with increased/decreased oxygen affinities, two mutant VHb’s for D44 (D44E and D44K) were generated and characterized using UV-visible/FTIR spectroscopy. Oxygen affinities of wild-type and mutant VHb’s were determined. These data suggest that position 44 may be a promising target for engineering oxygen affinity of VHb.
