| Abstract | Amac: Bu cal›flman›n amac› Nd:YAG lazerlerin farkl› frekans ve guclerde uygulanmas›n›n periapikal bolgede, ozellikle alveoler ke- mik dokuda, hucresel boyutta meydana getirebilecei deiflikliklerin hucre kultur yontemleri kullan›lak incelenmesidir. Gerec ve Yontem: Cal›flmada insan osteoblast benzeri devaml› hucre kulturu Saos-2 kullan›ld›. Nd:YAG lazer hucre kulturleri uze- rine temass›z flekilde 20-120 mj, 10-30 Hz ve 0.2-3.6 W ayarlar›nda 10 saniye sureyle uyguland›. Ayn› flekilde Nd:YAG lazerin reh- ber ›fl›¤› olarak kullan›lan He-Ne lazer, hucre kulturleri uzerine temass›z flekilde 10 saniye sureyle uyguland›. 7, 14 ve 21 gunluk in- kubasyon doneminden sonra hucresel aktivite Alkalen fosfataz (ALP) ve osteokalsin olcumleriyle deerlendirildi. Bulgular: ALP aktiviteleri karfl›laflt›r›ld›¤›nda 20 mj, 10 Hz, 10 saniye ve kontrol grubu aras›nda anlaml› bir fark bulunmad› (p>0.05). Dier gruplar aras›ndaki fark anlaml› bulundu (p<0.05). ALP ve osteokalsin ortalama deerlerine gore bir deerlendirme yap›ld›¤›n- da, lazerin at›m enerjisi ve enerji c›k›fl›n›n artt›r›lmas›n›n osteoblast hucre kulturleri uzerinde negatif bir etki oluflturduu goruldu. Sonuc: Eer at›m enerji miktar› ve enerji c›k›fl›, amaclanan tedavi icin yeterliyse en dufluk ayarlar› secmek doru yontem olacakt›r. Anahtar kelimeler: Nd:YAG lazer, He-Ne lazer, ALP, osteokalsin, hucre kulturu SUMMARY Objective: The aim of this study was to evaluate the effects of Nd:YAG laser usage on alveoler bone at celluler level by using cell culture methods. Material and Method: In this study we used Nd:YAG laser on human osteoblast like cell line Saos-2 in a non-contact mode for 10 seconds with 20-120 mj, 10-30 Hz, and 0.2-3.6 W parameters. He-Ne laser which is the guide light of Nd:YAG laser was used in a non-contact mode for 10 seconds. After 7, 14 and 21 day incubation period, cellular activity was assessed with Alcaline phosphatase (ALP) and osteocalcin mesuarements. Results: When ALP activities were compared, there was no statistically diffence between 20 mj, 10 Hz, 10 seconds and control gro- up. The difference between all other groups were also significant (p<0.05). When avarage ALP and osteocalcin activities were com- pared, increase in the pulse energy and power output had a negative effect on human osteoblast like cell cultures. Conclusion: It was concluded that; usage of the lowest parameters should be choosen if the pulse energy and power output was appro- priate for the suggested theraphy. |
