| Abstract | Ofloxacin is a second generation floroquinolon used as an antibacterial agent in the treatment of urinary tract infection, prostatitis, skin and respiratory tract infections. It is also recommended for the treatment of gonorrhea, urethritis, and cervicitis. Ofloxacin in tablet dosage form is available in the market; generics and trademarks. Drug quality inspection is required to ensure the drug arrives at the target site and provides a therapeutic effect. The objective of this study was to estimate ofloxacin in tablet dosage form using ultraviolet spectrophotometric method, whether it meets the requirement of a drug quality. Ofloxacin estimation in tablet dosage form was determined using ultraviolet spectrophotometric method at 297,5 nm and methanol as the solvent, and the results of analysis were validated statistically based on accuracy using the standard addition method, precision, limit of detection (LOD) and limit of quantitation (LOQ). The estimation of ofloxacin in Tariflox ® (Lapi) was found to be (101.467 ± 0.418)%; (100.975 ± 0.400)% in Ofloxacin (Novell); (104.21 ± 0.416)% in Ofloxacin (Indofarma); (98.428 ± 1.897)% in Danoflox ® (Dankos); (98.407 ± 1.533)% in Tarivid ® (Kalbe); and (104.418 ± 1.677)% in Akilen ® (Sanbe). The validation data showed that the ultraviolet spectrophotometric method can be used in the determination of ofloxacin in tablet dosage form as it was accurate and precise, and the limit of detection (LOD) and the limit of quantitation were 0.2878 μg/ml and 0.9595 μg/ml respectively. The results of analysis showed that ofloxacin in both generic tablet dosage form and branded tablet dosage form fit the required amount stated in the Farmakope Indonesia V (2014), which is not less than 90.0% and not more than 110.0% of the labeled amount. |
