| Abstract | The mixture of chloramphenicol and prednisolone is one of combination in cream supply. Determination of chloramphenicol and prednisolone in cream can not found in any monography, either on the Indonesia Pharmacopeia 5 edition (2014) or United States Pharmacopeia (USP) 30 edition (2007) so that i t required an analysis method that fulfill the test of validity in determining the mixture. The aim of this study is to determine of chloramphenicol and prednisolone in cream and to test the validation of derivative spectrophotometry method. The method of this research was done by purposive sampling to chloramphenicol and prednisolone mixture of the sample X and sample Y in cream content using derivative spectrophotometry with zero crossing technique and determination used ethanol absolute solution. Wavelengths analysis to determine of chloramphenicol and prednisolone in the second derivate is 227.60 nm for chloramphenicol and 292.80 nm for prednisolone. The result of this research is obtain that the chloramphenicol of sample X in cream each gram is (19.63-20.09) mg and sample Y in cream each gram is (19.4420.27) mg and prednisolone of sample X in cream each gram is (2.472.58) mg and sample Y in cream each gram is (2.50-2.62) mg. Based on this results of analysis determine the measure of chloramphenicol and prednisolon in cream supply chloramphenicol is fulfill and prednisolone is fulfill either, it requirements in USP 30 edition (2007) and Indonesia Pharmacopeia 5 edition (2014). The results of validation test on sample X, % recovery for chloramphenicol is 99.20%, relative standard deviation (RSD) = 1.72% and for prednisolone, % recovery = 101.42%, relative standard deviation (RSD) = 1.69%. Based on the results of research, that derivative spectrophotometry with zero crossing method fulfilled the requirements of accuracy and precision and can be used to determinate the content of chloramphenicol and prednisolone in cream. |
