RAHAYU and SITI NURJANAH Sensitivity is an important component in qualitative detection methods. The purpose of this study was to study the effect of pre-enrichment step in increasing the sensitivity of simplex PCR method on detecting Salmonella spp., S. Typhimurium, and S. Enteritidis and its application to chicken carcasses. Contamination of Salmonella spp. can be detected by simplex PCR method at 2.3 CFU / 25 grams for Salmonella spp. and S. Enteritidis, and 0.9 CFU / 25 gram for S. Typhimurium using pre-enrichment step on BPW media for 8 hours. Samples from supermarkets and traditional markets showed positive result of Salmonella spp., S. Typhimurium, and S. Enteritidis. Sample from supermarkets (n=6) showed the increase of positive results after getting pre-enrichment step from 83 to 100% for Salmonella spp., from 0 to 50% for S. Typhimurium, and from 50 to 83% for S. Enteritidis. Samples from traditional markets (n=6) showed the increase of positive results after getting pre-enrichment step from 60 to 100% for Salmonella spp., from 16 to 50% for S. Typhimurium, and there was no difference for S. Enteritidis (60%). |
