Supervised by SITI NURJANAH and WINIATI P. RAHAYU. Salmonella is one of the most common pathogenic bacteria that found in food, especially serotype S. Typhimurium and S. Enteritidis. Poultry meat especially chicken carcass is one of food material that easily contaminated by Salmonella. Rapid detection method for Salmonella using multiplex real-time Polymerase Chain Reaction (rt-PCR) is needed so that the detection is faster and more efficient. This study aims to develop Salmonella spp. rapid detection and serotyping methods using multiplex rtPCR. The research stages included DNA extraction, optimization of primary concentration, specificity test and limit of detection, creation of standard curves, and trials on chicken carcasses. The extraction method was cell lysis method. The purity of Salmonella DNA extraction results were between 1.84-1.93 and the concentration between 371.30-985.00 ng/μL. The primers used were InvA119 for Salmonella spp., STM523 for S. Typhimurium, and Prot6E for S. Enteritidis. The optimum primers concentration were 0.3 μM for InvA119, 0.6 μM for STM523, and 0.1 μM for Prot6E. Detection limits for all three target genes and multiplex were 0.00288, 0.02200, and 0.00218 ng/μL respectively and the number of cell were 1.9x10, 4.1x10 and 2.1x10 cells. Detection limit for multiplex was 0.00727 ng/μL but the concentrations of each Salmonella serovars were unknown so the lowest cell detected could not be calculated. The R standard curves ranged 0.7-0.8 and invalid for quantification. |
