| Abstract | In the present study, the inhibitory effect of arazyme on allergic inflammation was investigated by evaluating the alteration of cytokine production and expression of skin barrier proteins in immune and HaCaT human keratinocyte cells. THP1 human monocytic and EoL1 human eosinophilic cells were treated with Dermatophagoides pteronissinus extract (DpE). Monocyte chemotactic protein1 (MCP1)/CCL2, interleukin (IL)6 and IL8 increased following DpE treatment and arazyme significantly blocked the increase of MCP1, IL6 and IL8 expression in cell types. Secretion of MCP1, IL6 and IL8 induced by lipopolysaccharide in THP1 cells was also inhibited by arazyme treatment. Arazyme inhibited the secretion of IL6 and IL8 due to phorbol 12myristate 13acetate and calcium ionophores in human mast cells. Arazyme blocked the secretion of thymus and activationregulated chemokine (TARC)/CCL17, MCP1, IL6 and IL8 due to tumor necrosis factor (TNF ) and interferonγ (IFNγ) in HaCaT cells. TNF and IFNγ suppressed the expression of skin barrier proteins, including filaggrin, involucrin and loricrin. By contrast, arazyme increased the expression of filaggrin, involucrin and loricrin. These results may contribute to the development of a therapeutic drug for the treatment of allergic diseases, including atopic dermatitis. |

National Digital Library of India (NDLI) is a virtual repository of learning resources which is not just a repository with search/browse facilities but provides a host of services for the learner community. It is sponsored and mentored by Ministry of Education, Government of India, through its National Mission on Education through Information and Communication Technology (NMEICT). Filtered and federated searching is employed to facilitate focused searching so that learners can find the right resource with least effort and in minimum time. NDLI provides user group-specific services such as Examination Preparatory for School and College students and job aspirants. Services for Researchers and general learners are also provided. NDLI is designed to hold content of any language and provides interface support for 10 most widely used Indian languages. It is built to provide support for all academic levels including researchers and life-long learners, all disciplines, all popular forms of access devices and differently-abled learners. It is designed to enable people to learn and prepare from best practices from all over the world and to facilitate researchers to perform inter-linked exploration from multiple sources. It is developed, operated and maintained from Indian Institute of Technology Kharagpur.
