| Abstract | In the present work, a study has been carried out on the transmembrane protein p75, which is a neurotrophin receptor, which, among other functions described, is believed to exert a protective effect on cells under oxidative stress conditions. In order to evaluate that, HEK cells were exposed to oxidizing conditions by incubation with H2O2 and the lysates obtained were revealed by western blot. Thus it has been discovered that p75 forms dimers under oxidizing conditions, in which C257 cysteine appears to be involved by the formation of a disulfide bound. To evaluate the possible implication of another cysteine, C379, the intracellular fragment of p75 wt and with a mutation in this cysteine to alanine (C379A) has been purified and its behavior in solution has been evaluated, reaching the conclusion that, althought it is not essential for the formation of the dimer, it can have a function stabilizing them. Finally, the protective function of p75 was evaluated through flow cytometry, observing that the protein per se does not generate any protective effect, but that in the presence of the neurotrophin NGF and the receptor tyrosine kinase A (TrkA), it acts to enhance the pro-survival effect of TrkA. |
