| Abstract | Cocoa is a perennial tree; it is cultivated in humid tropical areas. It is a crop of great economic importance for Africa, Asia, and Latin America. The main problem of cocoa cultivation is conventional propagation methods since these are very laborious, need intensive labor and these have low productivity. Therefore, is necessary the application of biotechnological technique for cocoa propagation as an alternative to conventional methods. One of them is in vitro culture of plant tissues which is an efficient and low cost alternative for these purposes. The aim of this research was implement a protocol for obtaining calluses of CCN-51 cocoa (Theobroma cacao L.) with commercial interest. To achieve this aim, five treatments were carried out in callus induction phase using modified Driver and Kuniyuki (DKW) salts. By means of a parametric analysis of ANOVA variance, two variables were evaluated, time of callus formation (weeks) and percentage of callus transformation. In these analysis, no significant differences were found in any of two mentioned variables, however treatment 2 (T2) was considered as the best treatment about time of callus formation since it had the lowest register in relation to the study variable (3,37 weeks). As for percentage of callus transformation variable, treatment 4 (T4) was considered as the best treatment because it was presented the highest register regarding cited variable (99,99%). Because of this study, the culture medium of treatment T4 for callus induction of CCN-51 cocoa was chosen as the best treatment because with this was obtained the greatest amount of callus in this species which is convenient to agronomic level. |
