| Description | Telomeres have a 3' overhang of G-rich nucleotides that is recognized by the reverse transcriptase, telomerase, one of the most common telomere maintenance mechanisms (TMM). Since the misregulation of telomere length and maintenance mechanisms are implicated in many age-related diseases, telomere syndromes and cancer, telomeres and their maintenance mechanisms are excellent candidates for therapeutic intervention since they are critical in maintaining basic human health. Telomere length changes and engagement of TMM are implicated in many diseases, especially those that are associated with ageing. A premature ageing condition known as Dyskeratosis Congenita (DC) is disease that is marked by telomere shortening that is more rapid than the normal ageing-related telomere shortening and a decrease in telomerase activity. Both telomere length and alterations in TMM are associated with many types of cancer. TMM are typically inactive in somatic tissues but are often activated in cancers through exploitation of telomerase or Alternative Lengthening of Telomeres (ALT).
