| Abstract | The 3′ untranslated region (3′UTR) and the signal peptide sequence (SPS) are important factors affecting protein transcription and translation. To investigate the effect of the 3′UTR and SPS on the activity of expression products of Pseudomonas pseudoalcaligenes insecticidal protein gene (ppip), the ppip constructs containing 3′UTR or SPS were cloned by PCR and then transformed into tobacco. The insecticidal proteins obtained in the different ppip-transformed tobacco strains were analyzed by SDS–PAGE and verified by Western blot analysis. The insecticidal activity of crude proteins expressed by different ppip constructs was determined by calculating the average mortality of larvae. Crude protein expressed by ppip with the SPS caused mean mortality of second and third instar larvae of 18.34% whereas for those without the SPS mean mortality was 71.67%. Constructs containing the 3′UTR had no significant effect on the mortality of larvae. This first report about the effect of 3′UTR and SPS on the ppip product is helpful for effective genetic engineering of insecticidal plants. |
