| Abstract | In order to explore the T cell repertoire to myelin basic protein (BP) of both multiple sclerosis (MS) patients and healthy subjects (HS), we raised BP reactive T cell lines and clones in 8 MS patients and 5 HS. These T cell fines were isolated from the peripheral blood after in vitro triggering with human BP. The magnitude of the proliferative response to human BP was not significantly different between the two groups. In contrast, when analyzing the patterns of recognition of human BP versus heterologous BP (of known amino acid sequence), we observed that most T cell lines from MS patients showed a preferential response to human BP and four patients did not show any response at all to rat BP, monkey BP or both. This may indicate that the T cell repertoire to BP in MS patients differs from that of healthy subjects. Fine specificities of these T cell lines and clones are currently being investigated using synthetic peptides. |
