| Abstract | The genus Brucella comprises a group of Gram-negative bacteria pathogenic to mammals and are labeled as Class III in the hazard classification. There are many difficulties to identify them: the need of many media for inoculation, they are slow growing, and there is a need of immune-serum and phage typing. Brucella sp. is the principal bacteria responsible for acquired infection in laboratories. Other sources of infection are through the contact with infected animals and ingestion of raw milk and its derivatives. The search for rapid and safe methods to identify this bacteria on the level of species or biovars would decrease the risk of infection and accelerate its control. The aim of this work was to use a BOX-PCR technique for the identification of Brucella spp. using references and Brazilian strains. We analyzed 38 Brucella strains and 13 bacteria with are important to test the specificity of this technique. All Brucella strains were previously identified by classical microbiological identification and by genus-specific PCR. The BOX-PCR regrouped all Brucella sp. into one group between 100 and 74% similarity. Differences between strains could be observed by the presence or absence of some fragments. However, it was not possible to differenciate each species or biovar because of the differences. All Brucella strains showed common fragments among them and can be used for the characterization of the genus.
