| Abstract | Sex selection by flow cytometry/cell sorting involves the staining of spermatozoa with Hoechst 33342 in combination with the impact of a u.v. laser beam, two potentially mutagenic agents. A livestock breeding efficiency, phenotypic and cytogenetic study after insemination with spermatozoa stained with Hoechst 33342 and sorted spermatozoa of sows born from flow-sorted spermatozoa and sows born from untreated spermatozoa was performed. Firstly the effect of these agents on the reproductive performance on the first generation was evaluated. Cytogenetic study was performed from peripheral blood lymphocytes from piglets born after insemination with flow sorted or unsorted (control) spermatozoa of sows of the first generation. Cells were collected by centrifugation, subjected to hypotonic solution and fixed and dropped onto slides. Sister Chromatid Exchanges (SCEs) and chromosome aberrations ( CA:including chromosome and chromatid breaks) per cell were scored in 50 second division metaphase from each donor. Data were analyzed by ANOVA. Reproductive parameters and litters performance of all inseminations performed were also recorded in all groups. Farrowing rates were 40% and 81.2% (p 0.05) of SCE and CA frequencies were observed in piglets born from flow sorted spermatozoa, or from untreated sperm (second generation). The results indicate that no mutagenic effect on spermatozoa expressed as increase in the incidence of abnormalities in the resulting offspring and also as increase in SCEs and CA frequencies on lymphocytes from these individuals, was induced by the flow sorting procedure in the second generation of piglets born. |
