| Abstract | In the present work we describe the standardization of capture ELISA for the detection of T. cruzi antigens in urine and serum samples of experimentally infected Cavia porcellus specimens (guinea pigs), using polyclonal antibodies obtained from three species that were immunized with antigens of T. cruzi. The antibody pair was selected: IgY anti membrane protein from Gallus gallus domesticus (hen) as capture antibody (4 μg/ml) and IgG anti-TESA from Vicugna pacos (alpaca) as detection antibody (4 μg/ml), purified by affinity chromatography. In addition, the conjugate antillama was used at a dilution of 1/7500, blocking buffer composed of PBST plus 6% semi-skimmed milk and 1% BSA, and the substrate was ortho-phenylenediamine. The limit of detection of TESA antigen in artificially contaminated urine sample was 4 ng. The levels of antigenuria were positively related to high levels of parasitaemia. The methods used for the concentration of urines show no significant differences in the kinetics of detection of T. cruzi antigens by ELISA. Regarding the kinetics of detection of antigens in serum, significant differences were obtained between the samples of guinea-pig sera (Cavia porcellus) infected and healthy. The results show that this assay is important and can be used as a tool for the diagnosis of T. cruzi in humans. |
