Aim: To investigate the changes in tissue levels and release of neuropeptides in neuromas which form following peripheral nerve injury and the influence of morphine on the release. Methods: Neuromas were induced in anaesthetized rats by ligating and cutting the saphenous nerve. Neuropeptide tissue levels were measured in acetic acid extracts of 1, 3 and 5 week (w) old neuromas. The SP tissue levels were also measured after 24hr immersion of neuromas in saline containing morphine (10 pg/ml) in situ. To stimulate a release of the neuropeptides the neuromas were superfused in situ with Tyrode solution containing bradykinin (100 pg/ml). Superfusates and tissue extracts were assayed for SP and CGRP by radioimmunoassay. Results: Levels of the neuropeptides in the 3 w old neuromas were significantly reduced compared to levels in normal saphenous nerve. Levels returned to normal after 5 w. Morphine increases tissue levels of SP in 5 w old neuromas, possibly by inhibiting the basal release of SP. Morphine did not alter the levels of SP in the 3 w old neuroma. Superfusion of neuromas with bradykinin resulted in an increase of SP and CGRP in the superfusate. The amount of SP released following super-fusion with bradykinin varies with the age of the neuroma and is directly proportional to the SP tissue levels. Conclusions: The changes in neuropeptide levels and release may be an important factor in the pathophysiology of neuroma nerve fibre sprouts. (Supported by the DFG.) |
