| Abstract | El present treball ha estudiat el paper de la insulina i els factors de creixement tipus insulina (IGFs) en el creixement i metabolisme de la truita (Oncohrynchus mykiss) i l?orada(Spaurus aurata). Per assolir aquest objectiu s?han realitzat experiments in vivo en ambdues especies en les quals s?ha provocat una situacio de creixement compensatori. Mitjancant la tecnica de RT-PCR real time s?ha analitzat l?expressio dels principals reguladors miogenics (miogenina, myoD), aixi com dels FGFs, miostatina, IGF-I e IGF-II, i de les dues isoformes del receptor d?IGF-I, durant el creixement compensatori de la truita irisada. De la mateixa manera s?ha estudiat la variacio en els nivells de binding dels receptors d?insulina i d?IGF-I en la musculatura blanca durant aquest mateix fenomen. En l?orada s?ha estudiat el paper dels receptors d?insulina i d?IGF-I en la musculatura blanca, aixi com els valors plasmatics d?insulina, i d?IGF-I durant els diferents periodes experimentals, igualment, s?ha analitzat la mobilitzacio de les diferents reserves energetiques (lipid, proteina i glicogen) en el muscul blanc i el fetge. Per a completar els nostres estudis in vivo, s?ha analitzat mitjancant la tecnica de RT-PCR real time i per primera vegadaen peixos, el patro d?expressio dels receptors d?hormona de creixement (GHRs) en la musculatura blanca i el fetge de truita irisada, aixi com l?expressio dels IGFs en ambdos teixits durant la realimentacio de la truita irisada. Despres de l?obtencio de nombrosos i novedosos resultats in vivo, es va procedir a la posada a punt d?un cultiu de cellules musculars d?orada. En aquest sistema in vitro s?ha estudiat el paper de la insulina i dels IGFs en l?estimulacio de les diferents vies de transduccio de la senyal al llarg del cultiu. De la mateixa manera, s?ha estudiat el paper metabolic d?aquests peptids realitzant estudis de captacio de glucosa i d?alanina en els diferents dies del cutliu. Per a confirmar els efectes d?aquests peptids eren mediatitzats per la presencia de receptors especifics s?han realitzat estudis de binding per al receptor d?IGF-I en els diferents dies de cultiu i s?ha estudiat l?efecte de diferents inhibidors de les vies de transduccio de la senyal al llarg del cultiu. " SUMMARY: The aim of the present work is to study the role of insulin and insulin like growth factors (IGFs) on growth and metabolism processes in the rainbow trout (Oncohrynchus mykiss) and in the gilthead sea bream (Spaurus aurata). The goal of this thesis has been assessed by means of in vivo experiments in both species evoking a situation of compensatory growth. By means of RT-PCR real time the expression profile of several important muscle regulators (miogenin, myoD), IGFs (IGF-I and IGF-II), fibroblast like growth factors (FGFs), myostatin and IGF-I receptor isoforms (a and b), has been analyzed during this situation in the rainbow trout. In the same manner the binding levels for IGF-I receptor has been studied under this conditions in this species. We have performed similar experiments using the gilthead sea bream. We have measured changes on insulin and IGF-I receptors in the white skeletal muscle, plasma levels of insulin, IGF-I and free fatty acids. In the same manner we have correlated this plasma changes with variations in body energy storages mobilization (lipids and glycogen). Our studies in vivo have been concluded analyzing as a first time in fishes, the time course expression profile of growth hormone receptors (GHRs)in both liver and muscle during refeeding, together with the analysis of the expression profile of IGFs in the same organs. After all the approximations in vivo we have developed a primary muscle cell culture in the gilthead sea bream. We have studied the role of insulin and IGFs in the stimulation in the main cell signalling pathways (AKT and MAPK). In the same manner, we have studied the metabolic role of these peptides analyzing both glucose and amino acid uptake among the different days of the cell culture. In order to confirm that all this mechanisms are mediated by IGF-I receptors, binding studies have been performed throughout the cell culture and the effect of several transduction pathways inhibitors has been also investigated. " |
