Diabetes mellitus or hyperglycemic is a chronic metabolic disorder due to insulin dysfunction that resulting in high blood sugar levels. One antihyperglycemic therapy is by using α-glucosidase enzyme inhibitors which inhibit the absorption of glucose. Preliminary study showed that several species of Aquilaria are potential as an antihyperglicemic. In this study, ethanol extract of Aquilaria filaria (gaharu) leaves was examined as antihyperglicemic and the kinetics of antihyperglicemic mechanism through α-glucosidase enzyme inhibition based on in vitro tests was observed. The results showed that the ethanol extract of gaharu leaves was potential as an antihyperglycemic with the IC50 value of 26 ppm. The maximum inhibition value of 49% was observed in gaharu leaves ethanol extract in concentration of 25 ppm. Study of inhibition kinetics showed that gaharu leaves inhibit α-glucosidase enzyme through mixed inhibition mechanism indicated by a decrease in the Vmax value of 2.18 to 1.37 and an increase in Km value of 0.30 to 3.64. |
