| Abstract | LISTIA VIDYAWATI M MANURUNG. Inhibition Kinetics of Surian (Toona sinensis Roem.) Leaf Extracts toward α-Glucosidase. Supervised by SYAMSUL FALAH and SYAEFUDDIN. Toona sinensis Roem known as surian widely found in the Southeast Asia, particularly in Java, Indonesia. This plant is widely used as a herbal medicine in China, including for diabetes treatment. This study aims to determine and compare inhibitory activity of extracts leaves of surian against α-glucosidase and the inhibition kinetic parameters. Surian leaves extracts obtained by reflux in water at a temperature of 90 °C and maceration using ethanol 70%. The crude ethanol extract then fractionated by fractionation based on polarity, using nhexane, diethyl ether and ethyl acetate. Fraction of n-hexane showed strongest inhibitory activity with an IC50 value of 26.04 μg/mL, followed by the fraction of diethyl ether, ethyl acetate, ethanol 70% and water, the IC50 values for these extracts is 33.83 μg/mL, 45.18 μg/mL, 101.95 μg/mL, 211.79 μg/mL, respectively. Kinetics study showed that the leaves surian actively inhibits αglucosidase through non-competitive inhibition mechanism by reducing Vmax from 12.80 x 10 -3 mM/minute to 6.60 x 10 -3 mM/min and Km values were not significantly changed (0.1188 mM to 0.0871 mM). |
