| Abstract | Introduction: Brucellosis, a contagious disease of livestock, known as Maltese fever or melitococcus. It is an anthropozoonosis, due to bacteria of the genus Brucella. Its extension is global, with a predominance in the Mediterranean basin and the developing countries, where it still poses a real public health problem, and represents a significant economic cost. Prevention of the disease is essential, given its risk to human health and its significant economic impact. Objective: Construction of a DNA library by DNA extraction from blood taken from patients with brucellosis disease and resistant and also a design of primers framing the TNF alpha gene. Materials and methods: 21 samples taken, including 8 resistant and 13 patients, in the department of infectious diseases of the Tlemcen University Hospital. The extraction of the DNA is carried out using the NaCl technique. These DNAs are assayed by spectrophotometry at 260 nm and 280 nm. Specific primers surrounding the TNF alpha gene are designed by the Primer-Blast tool. Results: A total of 13 DNA is extracted from patients, and 8 DNA from resistant subjects. The dosage of these DNAs showed 21 DNAs specifically contaminated by proteins. |
