Abstract	t Tissuee factor (TF) is a transmembrane protein, which is essential for initiation of thee coagulation cascade. TF has been reported to play an important role in the progressionn of endotoxin (lipopolysaccharide, LPS)-mediated sepsis, being inducedd in numerous tissues, such as kidney, spleen and lung. We developed andd validated a rabbit anti-murine TF peptide antiserum to localize TF protein in a murinee sepsis model. TF protein distribution was compared to localization of TF mRNAA and fibrin deposits, the ultimate resultant of procoagulant TF activity. While evidentt LPS-mediated TF mRNA induction was observed in the tubular area at thee cortico-medullar junction in the kidney, there was no increase of TF protein afterr LPS administration. In the spleen, however, TF mRNA is induced in the interfollicularr region upon LPS injection, corresponding to increased TF protein in thee same area. The clusters of TF-protein positive cells in the spleen are predominantlyy granulocytes, suggesting that the granulocytes synthesize TF mRNAA and protein. Based on this observation, we hypothesize that these spleenderivedd cells transport TF to other locations to initiate fibrin formation, presumably too support the maintenance of tissue integrity.
