| Abstract | This study was conducted to examine genetic variations of the growth hormone gene in the four pig breeds (Landrace, Large White, Duroc, and Meishan) by analysis composed of Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Genomic DNAs were prepared from the blood samples of 163 animals consisting of 78 Landrace, 29 Large White, 17 Duroc, and 39 Meishan. The PCR reaction was performed with 50 ng of genomic DNA with the Taq DNA polymerase for 40 cycles of amplification (denaturation at 94•Ž for 30 sec, annealing at 60•Ž for 1 min, extension at 72•Ž for 30 sec). Synthetic oligonucleotides were used as primers : forward, 5'GAGGCCAAGTTTTAAATGTCCCTG-3' ; reverse, 5'TTGGGAATCCCTTCAGTTTACAC-3'. After amplification, 422 bp of PCR product was digested with four endonucleases (Apal, Cfol, Ddel and Mspl), and separated by polyacrylamide gel electrophoresis. Genotypes of 163 animals were determined and then the genetic variabilities were calculated from their allelic frequencies. Some differences of allelic frequencies were observed among the 4 breeds. At the Cfol site, Landrace pigs have C1, C3 and C4 alleles, Duroc have C3 and C4, Meishan have C1 and C2, and Large White have all alleles. No polymorphisms were detected at the Apal, Ddel and Mspl sites in Meishan pigs, whereas other three breeds have both 2 alleles. Comparing genetic variabilities among 4 breeds, the lowest (0.189) and highest (0.913) average Homogeneity Index (HI) were observed in the Landrace and Meishan pigs, respectively. The values of HI in the Large White and Duroc pigs are 0.287 and 0.405, respectively. The other indices of genetic variabilities were similar to these results among 4 breeds. However, genetic variability at the Mspl site in Large White pigs was extremely low (HI, 0.932), whereas HI at the other 3 sites were higher than ones in Landrace pigs (0.043-0.113). 15 小林 ら:ブ タ成長 ホルモ ン遺伝子 における遺伝的多型性 |
