This change has been driven by technological advances in biological science, rapid follow-up activities in chemistry, and powerful computer techniques. Of these, structure-based drug design (SBDD) and combinatorial chemistry (CC) have proved to be successful methodologies. Knowledge of the three-dimensional (3D) structure of a biological target can help to identify a lead compound and subsequently guide its optimization. Combinatorial chemistry, on the other hand, is a synthetic strategy whereby molecular diversity can be rapidly reproduced in the laboratory with the specific aim of targeting a biological system.
