| Abstract | A novel simple, fast qualitative method for detection and differentiation of PVY strains in potatoes by the LightCycler technology is described. Fluorescent-labeled probe designed to contain variable degree of homology with a ‘target sequence’ of several known reference PVY strains was annealed to PCR products of these viruses followed by a graded melting analysis. The specific characteristics of the melting curves enable the detection, distinction and differentiation of each of following four known PVY strains O-FL, O-RB, N and NTN, in a single reaction obviating the need for size or nucleotide sequence analyses. In addition, it was demonstrated that virus extracts from plants infected with more than one strain can be resolved using this procedure. The melting-curves of extracts from 55 market size tubers harvested randomly from commercial fields and compared with those of the reference virus strains, revealed the presence of O-FL, O-RB and NTN strains of PVY.Se describe un método novedoso, simple, cualitativo, rápido, para la detección y diferenciación de variantes de PVY en papa por la tecnología de LightCycler. Se templó una sonda etiquetada fluorescente designada para contener un grado variable de homología con una “secuencia-objetivo” de varias líneas de PVY conocidas como referencia, para productos de PCR de estos virus, seguida por un análisis de graduación de disolución. Las características específicas de las curvas de disolución habilitan la detección, distinción y diferenciación de cada una de las siguientes cuatro variantes conocidas de PVY, O-FL, O-RB, N y NTN, en una sola reacción, obviando la necesidad de análisis del tamaño o de secuencia de nucleótidos. Además, se demostró que los extractos de virus de plantas infectadas con más de una variante se pueden determinar usando este procedimiento. Las curvas de fusión de extractos de 55 tubérculos de tamaño comercial cosechados al azar de campos comerciales y comparados con los de las variantes de los virus de referencia, revelaron la presencia de las variantes de PVY O-FL, O-RB y NTN. |

National Digital Library of India (NDLI) is a virtual repository of learning resources which is not just a repository with search/browse facilities but provides a host of services for the learner community. It is sponsored and mentored by Ministry of Education, Government of India, through its National Mission on Education through Information and Communication Technology (NMEICT). It is developed, operated and maintained from Indian Institute of Technology Kharagpur.
