NDLI: Silencing of c-myc gene expression using enzymatically and chemically synthesized siRNAs

Abstract	Synthetic small interfering RNAs (siRNAs) are of great interest as promising agents for targeted downregulation of gene expression. A study was made of the silencing of c-myc in KB-3-1 cells by siRNAs targeted to region 1452-1470 of the third exon of the c-myc mRNA and the conserved sequence of the second exon of the c-myc (697–715) and N-myc (302–320) mRNAs. To obtain siRNAs enzymatically, a scheme was used that included dsDNA template synthesis with the Klenow fragment, in vitro transcription of each of the siRNA strands, their subsequent hybridization, and elimination of the leader sequences with RNase T1. The enzymatically synthesized siRNAs decreased the c-myc mRNA level more efficiently than chemically obtained siRNAs of the same sequence. Silencing of c-myc by siRNAs inhibited or even completely arrested proliferation of epidermoid carcinoma cells. The inhibition of cell proliferation correlated with the inhibition of c-myc expression.
