| Abstract | Light Sheet Microscopy has been shown to be a powerful tool for the life sciences, in particular for developmental biology [1]. By illuminating the sample with a thin sheet of light, optical sectioning is achieved and since no out-of-focus light is generated, photo bleaching is minimized. Selective Plane Illumination Microscopy (SPIM, [2]) has become particularly popular, due to its unique three dimensional imaging capabilities such as rotation of the sample and multi-view fusion [3]. Although various advanced light sheet designs are being developed in research labs around the world, the basic SPIM technology is relatively simple and is of such great interest to developmental and cell biologists, that we believe every lab should have one. To that end we have developed an Open Access SPIM system (OpenSPIM). The microscope is easy to assemble, modular, comparably inexpensive and has a small footprint. Plans, part lists and step-by-step assembly instructions are available on our wiki site [4]. We believe that OpenSPIM will make light sheet microscopy accessible to a wider community including scientists without profound knowledge in microscopy. Importantly, the microscope is controlled by an Open Source program extending μManager [5] on the Fiji [6] platform. In Fiji, the data acquired using OpenSPIM can be directly processed and analyzed with dedicated plugins [3]. In this way we combine the ideas of Open Software and Hardware presenting a unified platform for further collaborative development of the SPIM technology and its applications. |
