Thesis (PhD) University of Brasília, Brasília, 2019. Infections caused by Gram-negative bacteria have been considered one of the major health problems worldwide. Gram-negative bacteria such as Escherichia coli and Pseudomonas aeruginosa are considered priorities for the search for new treatments according to a priority list published by WHO. The WHO also reports the importance of the search for new strategies for the treatment of these diseases. Thus, antimicrobial peptides may be an alternative for the treatment of these pathologies The antimicrobial peptides computer guided design has been used in the last decades to yield novel molecules. Among those methods, the Joker algorithm was used to design the peptide PaDBS1R6. In summary, this study aims to evaluate the in vitro and in vivo antibacterial activities of PaDBS1R6, along with the characterization of peptide interaction to target membranes and also the peptide structure investigation in contact with mimetic vesicles. Here we demonstrate that this peptide exhibited selective antimicrobial activity against Gram-negative bacteria. In addition, experiments with lipid vesicles showed that PaDBS1R6 structural arrangement transits from random coil to α-helix in the presence of negatively charged lipids as characterized by circular dichroism (CD) and nuclear magnetic resonance (NMR). Thus, the PaDBS1R6 peptide seems to be a candidate for Gram-negative bacteria healthcare-associated infections treatment and can also be used as a template for new antimicrobial agents. |
