Patogenicidade à cebola (Allium cepa L.) e análise da diversidade genética de isolados de Colletotrichum gloeosporióides do Estado de Pernambuco, Brasil, por RAPD e região ITS do rDNA

| Abstract | The leal anthracnose or evil-give-seven-turns of onion, caused by Colletotrichum gloeosporioides stands out as one of the main diseases in areas of production of the onion in the State of Pernambuco. This holated from amion work aimed to study the pathogenicity of C. gloeosporioides and to analyze the genetic variability through RAPD and the region ITS of the rDNA. Sixteen stram were used 11from different substrata and hosts and 5 from lesion C. gloeosporioides isolated of onions of different regions of Pernambuco and one from Amazonas. The pathogenicity test was accomplished in seedlings and bulbilhos of the onion. Four strain esolated from onion, were from most aggresivein both test. In the analyses of RAPD seven primers of arbitray sequences were used. The amplification products were separate in agarose gel to 1,4% and showed polymorphics bands that allowed to evaluate the genetic distance among the 15 strais. Which were classified in four groups. The products of amplification of the loci ITS1-5. 8 S-ITS2 of the rDNA with the primers ITS1 and ITS4 also presented polymorphism and were digested with three restriction enzymes Dra l, Hae III and Msp I. The last two just showed genetic variations among the isolated ones. Only the last two were udeful differentiate the strain from Amazonas from those al Pernambuco. The results obtained with the molecular markers they didn't show relationship with the pathogenicity degree of the strains onion. |
