| Abstract | The objective of the present work was to compare the anatomy of the type 1 primary callus (with no persistent growth), type 2 primary callus (with persistent growth), and embryogenic callus derived from Coffea arabica leaf explant, 'Catuaí Vermelho' cultivar. For this purpose the three types of calli were fixed in FAA solution (formalin, acetic acid, ethanol), dehydrated in an increasing ethanolic series, infiltrated by hystoresin and sectioned with the rotary microtome. The sections were stained with toluidine blue, and the results registered by a photomicroscope with image capture system. Cells from primary calli were bigger than embryogenic cells. The cells from type 1 primary calli present parenchymatic characteristics with thicker cell walls, while those from type 2 primary calli have bigger intercellular spaces. The embryogenic callus showed well defined regions with smaller isodiametric cells, with dense cytoplasm, prominent nuclei and intense cell division. The linear arrangement of embryogenic cells in some regions of the embryogenic calli may suggest the cell organization toward embryo regeneration. The present analysis has clarified the morphological differences between the three types of calli. |
