| Abstract | The Anti-psychotic agent used for the present study is Olanzapine. Olanzapine is an atypical antipsychotic drug of the thienobenzodiazepine class for treatment of schizophrenia and to control manic episodes of bipolar disorder. Olanzapine is well absorbed from GIT and its half-life ranges from 21-54 hours. So, the present investigation is concerned with the increase in patient compliance, fast onset of action and increase dissolution by its faster disintegration. Drug-polymer compatibility studies were done by FTIR and DSC, which gave conformation about their purity of the drug and showed no chemical interaction between drug and selected polymers. A Simplex-Lattice design was applied to optimize the effect of three factors, i.e. super-disintegrants like crospovidone, croscarmellose sodium and sodium starch glycolate. It was observed that the responses like disintegration time, hardness and friability were affected by all the factors. The prepared tablets were evaluated for appearance, hardness, friability, disintegration time, wetting time, water absorption ratio, estimation of drug content and in vitro drug release studies. All the formulations had disintegration time < 40 s and release maximum amount of drug in 10 min. From among all the developed formulations, as formulation F9 disintegrates in 16.6 s and release the 99% of drug in 10 min. F9 was found to be stable during stability studies according to ICH guidelines 40 ± 2 °C / 75 ± 5 % RH for six months. F9 satisfied physicochemical parameters, disintegration time, wetting time and in vitro drug release profile requirements for a fast dissolving tablet. |
