| Abstract | The purpose of this study was to evaluate the effect of different culture media on in vitro production (IVP), lipid content and rate of apoptosis of crossbred bovine embryos, using the modified synthetic oviduct fluid (mSOF) medium supplemented with polyvinyl-alcohol (PVA), bovine serum albumin (BSA) and fetal calf serum (FCS). For that, ovaries from slaughtered females were collected, the cumulus-oocyte complexes (COCs) were matured, fertilized and then the embryos were cultured in vitro in the different systems: (1) mSOFaaci+PVA (0.1mg/mL from day 1 (D1) to day (D8), (2) mSOFaaci+BSA (6mg/mL) from D1 to D8, (3) mSOFaaci+BSA + 5% FCS from D6 to D8 and (4) mSOFaaci+BSA+ 1% FCS from D1 to D8. The results showed no significant differences (P<0.05) among the rates of blastocysts at D8 produced by treatments whit PVA, BSA, BSA + 5% FCS (16.5; 19 and 15.6%, respectively), but there were differences between treatment whit PVA y BSA + 1% FCS (11.6%). For the lipidic content, treatments whit PVA and BSA have fewer lipidic droplets and mostly small drops, while with BSA + 1 and 5% FCS the presence of large drops was higher compared to the other treatments (P<0.05). Similarly apoptotic index increased by the presence of serum in the culture medium. In conclusion, the culture system with PVA is capable of supporting the crossbred embryos development. The serum-free medium used in this work to produce in vitro bovine embryos should be further improved to meet the requirements and demand at a large-scale. |
