Fifty-one populations of Meloidogyne spp. obtained in a root-knot nematode survey in grape (Vitis spp.) orchards in Rio Grande do Sul and Santa Catarina states, were characterized biochemically by esterase phenotype (Est). In order to evaluate the reaction of resistance of four tropical grape rootstocks, seven temperate rootstocks and two top cultivars to Meloidogyne incognita, M. arenaria, M. javanica and M. enterolobii, an assay was carried out at greenhouse conditions The resistance levels were characterized by the reproduction factor (RF) of each tested nematode species. The effect of the temperature in the hatching of M. ethiopica second-stage juveniles (J2) was investigated in a in vitro bioassay; An histopathological study in roots of grape rootstock resistant ('1103 Paulsen') and susceptible ('106-8 Ripária do Traviú') to M. ethiopica were also carried out where the resistance mechanisms were examinated microscopically. Fifty-five percent of grape samples presented the root-knot nematode and five Meloidogyne esterase phenotypes were characterized. The species. M. javanica, was the most frequent in grape orchards. In Rio Grande do Sul state were detected 34 populations of M. javanica Est J3 (Rm: 1.00, 1.23, 1.40) and Est. J2 (Rm: 1.00, 1.24), corresponding to 66.66% and 3.92% of the samples analyzed, respectively. Also, eight populations of M. arenaria Est A2 (Rm: 1.20, 1.29), five populations of M. incognita Est I2 (Rm: 1.05, 1.10), and two populations presenting the phenotype Est A3 (Rm: 1.14, 1.20, 1.29) of M. morocciensis, were detected in 15.69%, 9.8% and 3.92% of samples, respectively.
