| Abstract | Diclofenac is the most commonly prescribed non-steroidal antiinflammatory drugs (NSAIDs) that often to treat pain, fever, and inflammation, however the use of diclofenac in long term can induce peptic ulcer. The design of pro-drug is one of strategies to overcome this problem. Pro-drug is a pharmacologically inactive substance transformed by human system become an active substance with chemical or metabolism reactions. Pro-drug is designed to protect free acid group in a NSAIDs molecule thus protecting gastrointestinal tract from local irritation. In this study, the pro-drug is a propyl diclofenac. The purpose of this study are to find out esterification, to know the purity and the characteristics of propyl diclofenac. Propyl diclofenac was made by esterification of diclofenac acid with dry propanol and H2SO4(p) as a catalyst. Diclofenac acid was obtained by potassium diclofenac hydrolysis with HCL 2N that is characterized by FT-IR test. The purity of the result of propyl diclofenac synthesis was indicated by melting point test and then analyzed using FT-IR and GC-MS. FT-IR result of diclofenac acid showed that there were functional group changing became OH of carboxylic in 2993.52 cm, 2885.51 cm, 2681.05 cm, and 2576.90 cm. The result of propyl diclofenac melting point test were 120 124C. The result of FT-IR test showed there had changing of O–H functional group became CH2CH3 in 1496.76 cm and 1450.47 cm. The result of GC-MS showed that propyl diclofenac had been formed. The conclusion of this study showed that esterification of diclofenac acid could be done but a pure propyl diclofenac was still not obtained yet and the characteristic of propyl diclofenac could be obtained using FT-IR and GC-MS. |
