| Abstract | Different extracts from Punjab Haldi1 and Punjab Haldi 2 were prepared by three extraction procedures i.e. conventional, ultrasonic assisted extraction and soxhlet extraction methods using different solvents (hexane, benzene, isopropyl alcohol, ethyl acetate and acetone). Soxhlet extraction method was found to be best giving maximum total phenolic content for isopropyl alcohol extract of Punjab Haldi1 and Punjab Haldi 2 with values 70.37 and 39.63mg GAE/g DW, respectively. The yields of curcumin isolated from Punjab Haldi1 and Punjab Haldi 2 were found to be 22.35 and 29.75mg/g turmeric powder, respectively. Acetylated and methylated derivatives of isolated curcumin were prepared and identified by TLC, melting point, FT-IR and H NMR data. All the extracts of Punjab Haldi1 and Punjab Haldi 2 were investigated for their 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity using 3,5-di-tert-butyl-4-hydroxytoluene (BHT) as a standard and antibacterial activity by disc plate method against Bacillus aryabhattai, Acinetobacter sp. and Klebsiella sp. using tetracycline as standard. Among all the extracts, isopropyl alcohol extract showed highest antioxidant and antibacterial activities. From the isolated compound and its derivatives, curcumin was found to be more effective than its derivatives. |
