Description	Inhalation therapy is gaining importance in the therapy of a number of pulmonary diseases. This site-speciﬁc targeted delivery aims to achieve high local activity with reduced systemic side effects. Although numerous review articles have described the clinical efﬁcacy of these inhalation agents [1-8], detailed reviews on the underlying pharmacokinetics and their relevance for targeted drug therapy are rare [3, 9-11]. This chapter evaluates the importance of pharmacokinetic factors for pulmonary targeting and reviews modern pulmonary drug delivery approaches to achieving pulmonary targeting.
