| Description | Journal: Medical mycology A method is described for the isolation of undergraded RNA from yeast-like (Y) and mycelial (M) forms of Paracoccidioides brasiliensis. The method is a combination of various known techniques. Cosedimentation in a sucrose gradient of Y form RNA labelled with$ ^{14}$C-uracil and M form RNA labelled with$ ^{3}$H-uracil showed identical sedimentation behaviour of the 3 major types of RNA. However, one of these types of RNA (light ribosomal RNA) is comparatively more abundant in the M form than in the Y form of P. brasiliensis. In addition to the 3 major sedimenting types of RNA, many other intermediate species of RNA were detected in both Y and M forms by electrophoretic analyses in polyacrylamide gel. A two-band difference was found between electrophoretic patterns of RNA sedimentation samples obtained from Y and M forms. |
