| Description | In Chapter 4, we mainly considered the fixed sample size design, in which there is no interim analysis. As we alluded to in Chapter 1, however, clinical trials often require periodic monitoring and data analyses at interim stages, while patients are continuously enrolled. This monitoring and analysis is especially necessary for trials that involve life-threatening diseases, potentially toxic therapies, long-term follow-ups, or large sample sizes. In this chapter and in Chapter 7, we examine sequential designs and methods for clinical trials with interim stages, with an illustration using oncology trials.
