| Description | The Janus kinase (JAK)–signal transducer and activator of transcription (STAT) pathway is a key signaling pathway for a diverse array of stimuli, including a number of growth factors and cytokines. The JAK family of kinases include JAK1, JAK2, JAK3, and tyrosine kinase (Tyk2). The STAT family is comprised of STAT1, STAT2, STAT3, STAT4, STAT5A, STAT5B, and STAT6. Pharmacological approaches as well as genetically altered JAK and STAT mice have yielded important insights into JAK-STAT signaling in cardiovascular health and disease. The goal of this chapter is to highlight the role of JAK-STAT signaling in the cardiovascular system in aging, atherosclerosis, diabetes, heart failure, and hypertension. Specific emphasis is placed on the roles of JAK1 and JAK2 as well as STAT1 and STAT3, since they account for the majority of JAK and STAT expression in the cardiovascular system. Molecular pathways that promote and inhibit JAK-STAT signaling as well as some of the specific ligand and receptor pathways that utilize JAK-STAT will be discussed, including the cellular effects associated with chronic JAK-STAT activation.
