| Abstract | Transcription by RNA polymerase can stimulate localized DNAsupercoiling in Escherichia coli. Invivo, there is extensive experimental support for a“twin-domain” model in which positive DNA supercoils aregenerated ahead of a translocating RNA polymerase complex and negativesupercoils are formed behind it. Negative supercoils accumulatein the template DNA because the positive supercoils are preferentiallyremoved by cellular topoisomerase action. Yet, in vitro,clear and convincing support for the twin-domain mechanism has beenlacking. In this article, we reconcile this inconsistency by showingthat, in a defined in vitro system with plasmid DNAtemplates, a variety of sequence-specific DNA-binding proteins, such asthe bacteriophage λ O replication initiator or the E.coli lactose or galactose repressors, strikingly stimulatetranscription-coupled DNA supercoiling. We demonstrate further thatthis stimulation requires the presence in the DNA template of arecognition sequence for the relevant DNA-binding protein and dependson the production of long RNA chains by an RNA polymerase. Our data aremost consistent with a model in which specific DNA-binding proteinsfacilitate a twin-domain mechanism to enhance DNA supercoiling duringtranscription. More precisely, we suggest that some nucleoproteincomplexes, perhaps those that contain sharply bent DNA, can formbarriers that impede the diffusion and merger of independentchromosomal supercoil domains. Localization of DNA supercoilsby nucleoprotein complexes may serve as a generalmechanism for modulating DNA transactions that are sensitive toDNA superhelicity. |

National Digital Library of India (NDLI) is a virtual repository of learning resources which is not just a repository with search/browse facilities but provides a host of services for the learner community. It is sponsored and mentored by Ministry of Education, Government of India, through its National Mission on Education through Information and Communication Technology (NMEICT). Filtered and federated searching is employed to facilitate focused searching so that learners can find the right resource with least effort and in minimum time. NDLI provides user group-specific services such as Examination Preparatory for School and College students and job aspirants. Services for Researchers and general learners are also provided. NDLI is designed to hold content of any language and provides interface support for 10 most widely used Indian languages. It is built to provide support for all academic levels including researchers and life-long learners, all disciplines, all popular forms of access devices and differently-abled learners. It is designed to enable people to learn and prepare from best practices from all over the world and to facilitate researchers to perform inter-linked exploration from multiple sources. It is developed, operated and maintained from Indian Institute of Technology Kharagpur.
